Project Success of the Department of Organic Chemistry, Department of Chemistry
A project entitled “Design, Synthesis, and Selective Anticancer Activity of EGFR/HER2 Targeted Next Generation Inhibitors for the Treatment of Lung Cancer”, led by Prof. Dr. Amaç Fatih TUYUN, faculty member of the Division of Organic Chemistry, Department of Chemistry, Faculty of Science, Istanbul University, has been awarded for funding under the TÜBİTAK 1001 – Scientific and Technological Research Projects Support Program.
Assoc. Prof. Dr. Nilüfer BAYRAK from the Division of Organic Chemistry, Department of Chemistry, Faculty of Science, Istanbul University, and Assoc. Prof. Dr. Ayşe Tarbın JANNUZZI from the Division of Pharmaceutical Toxicology, Department of Pharmaceutical Sciences, Faculty of Pharmacy, Istanbul University, are participating in the project as researchers. Additionally, one master’s student and one doctoral student will take part in the project as research fellows.
The project aims to develop novel lead compounds with high therapeutic potential against lung cancer through a rational, mechanism-oriented approach based on scientific evidence. Building upon the research team’s previous findings that certain molecules with amino groups and significant core structures exhibit potent and selective cytotoxic activity against specific cancer cell lines, a library of newly designed heterocyclic amino-containing molecules will be synthesized. The synthesized compounds will be evaluated for their cytotoxic effects against lung cancer cell lines (A549 and PC-9) and healthy lung epithelial cells, and compared with a clinically used kinase inhibitor. Selected compounds with high efficacy will undergo detailed investigations into their effects on the cell cycle, apoptotic/necrotic activity, kinase inhibition (EGFR and HER2), and downstream signaling pathways (MAPK/ERK and PI3K/AKT). Furthermore, their structural and interaction properties will be analyzed through ADMET profiling and molecular docking studies.
The expected outcome of the project is the discovery of new EGFR/HER2 inhibitors that could serve as candidates for preclinical or early-phase clinical studies, contribute to drug development efforts, and identify compounds eligible for patent applications.
